Determining Protein Sequence

The primary structure of protein or peptide can be determined by several ways. One of these is by taking advantage of the selective cleavage of the enzymes (like trypsin and chymotrypsin) or chemicals (like cyanogen bromide).Β 

The diagram below summarizes the cleavage properties of these enzymes/chemicals.Β 

As an example of using these components, shown below is the cleavage of peptide:

From knowing these cleavage properties, we can then determine the sequence of the amino acids in a protein.Β
